CyTOF®质谱流式细胞术——The Scientist杂志评选出的2011年十大创新技术。

CyTOF质谱流式细胞术,采用金属元素标记物(通常是金属元素标记的特异抗体或者染料)标记或识别细胞表面和内部的信号分子,然后用流式细胞原理分离单个细胞,再用感应耦合等离子质谱(ICP-MS)观察单个细胞的原子质量谱,最后将原子质量谱的数据转换为细胞表面和内部的信号分子数据,并通过专业分析软件对获得的数据进行分析,从而实现对细胞表型和信号网络的精细观察。

这项研究通过这个可以称为大量细胞计数法的方法,观察了人类骨髓产生的不同形态细胞中及表面的34种物质,不但能正确归类10多种不同类型的免疫细胞,还能观察到各类免疫细胞的内部变化,从而预知可能发生的变化。这将有助于更快更广泛的测量处方药对人体细胞的反应及功效,提前发现细胞病变,研发出针对个人的治疗药物。
